Description

A colorimetric Sandwich ELISA from Assay Genie designed for the quantitative detection of Rat Lactoylglutathione lyase (Glo1), an enzyme that catalyses the conversion of hemimercaptal, formed from methylglyoxal and glutathione, to S-lactoylglutathione. GLO1 is involved in the regulation of TNF-induced transcriptional activity of NF-kappa-B, and dysregulation of the enzyme has been implicated in diabetes, neurodegenerative disorders, and cancer, making the kit applicable to research into disease mechanisms and potential therapeutic interventions in these areas.

The assay offers a detection range of 0.156–10 ng/mL with a sensitivity of 0.084 ng/mL. Reported precision includes an intra-assay CV of 6.9% and an inter-assay CV of 8.8%. Linearity was evaluated in serum, EDTA plasma, and heparin plasma (N=5) at dilutions of 1:2, 1:4, 1:8, and 1:16, yielding ranges of 84–118% for serum, 100–127% for EDTA plasma, and 92–117% for heparin plasma. Recovery averaged 103% (range 97–109%) in serum and 105% (range 99–111%) in plasma. The kit is validated for serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids, with specificity for natural and recombinant rat Lactoylglutathione lyase. The target is a homodimer with a molecular weight of 20,820 Da, associated with UniProt Q6P7Q4, and is listed for cancer research applications. For research use only.
