Built around a two-antibody capture-and-detection architecture, this Assay Genie kit provides quantitative measurement of lactoferrin in rat-derived research samples across 96 individual assay positions. Lactoferrin is an approximately 80 kDa iron-binding glycoprotein of the transferrin family, secreted by neutrophils and exocrine glands, and serves as a biomarker in studies of mucosal immunity, iron homeostasis, neonatal nutrition, gut barrier function, and inflammatory responses in rodent models.

The microplate arrives pre-coated with a monoclonal capture antibody specific for rat lactoferrin. Reconstituted standards and rat-derived samples such as serum or plasma, as well as milk, saliva, or tissue homogenate are pipetted into the wells, where lactoferrin binds the immobilized antibody. After washing, a biotinylated secondary antibody is added, followed by streptavidin-conjugated horseradish peroxidase and a chromogenic substrate. Absorbance is read on a standard plate reader, and sample concentrations are interpolated from the standard curve.

Reagents supplied with the kit are sufficient for one full microplate run and typically include the pre-coated plate, lyophilized recombinant lactoferrin standard, biotinylated detection antibody, streptavidin-HRP conjugate, substrate solution, concentrated wash buffer, diluent, and stop reagent. Researchers should supply their own precision pipettes, multichannel pipettor, plate reader, distilled or deionized water, and appropriate sample collection tubes.

The procedure is intended for in vitro research use only and is validated for rat samples. Applications include studies of iron metabolism disorders, antimicrobial peptide research, inflammatory bowel disease models, and characterization of lactoferrin expression in lactating mammary tissue or neutrophil-rich exudates. Standard curve fitting and quality control should follow the protocol booklet supplied with each lot.
