Rat Klotho ELISA Kit

A highly reliable and sensitive assay from Assay Genie, this sandwich ELISA kit is designed for the precise quantification of klotho in rat samples. It uses a Sandwich ELISA, Double Antibody format with reactivity specific to rat, and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit offers high specificity and accuracy with consistent, reproducible results, supporting research into klotho — a protein that plays a crucial role in regulating aging and age-related diseases such as kidney disease, diabetes, and cardiovascular disorders. Klotho acts as a co-receptor for fibroblast growth factor receptors and helps maintain phosphate and calcium homeostasis, making this assay a useful tool for investigating physiological and pathological processes and potential therapeutic targets.

Sensitivity is 46.875 pg/mL with a working range of 78.125–5000 pg/mL. Linearity across dilutions of 1:2, 1:4 and 1:8 was demonstrated in serum (85–105%, 84–101%, 81–97%), EDTA plasma (86–105%, 88–100%, 87–96%) and heparin plasma (85–104%, 82–100%, 83–98%). Recovery ranges were 92–103% in serum (average 97%), 85–101% in EDTA plasma (average 94%) and 90–100% in heparin plasma (average 95%). The protocol includes primary incubation at 37 °C for 90 minutes, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution. Reagents should be stored at 2–8 °C for 12 months.
