Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of Kidney Injury Molecule-1 (KIM-1), also known as T cell immunoglobulin mucin-1 (TIM-1), in rat samples. KIM-1/TIM-1 is a well-established biomarker for kidney injury and is involved in various renal diseases, making it a valuable target for research in nephrology. The kit uses a double-antibody sandwich format with colorimetric detection and is intended for use with rat models across preclinical and translational research applications.

Performance specifications include a sensitivity of 37.5 pg/mL and a measurable range of 62.5-4000 pg/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Recovery in serum averages 91% (range 86-98%), in EDTA plasma averages 97% (range 93-102%), and in heparin plasma averages 93% (range 87-101%). Linearity in serum ranges from 80-104% across 1:2 to 1:8 dilutions, with comparable performance reported in EDTA and heparin plasma matrices. The kit should be stored at 2-8°C and is stable for 12 months under those conditions.

The assay workflow proceeds through six steps: equilibration of reagents and TMB substrate, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10-20 minutes, and stop reaction with absorbance measurement at 450 nm. Sample preparation protocols are provided for serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate.
