The assay procedure involves equilibrating reagents and TMB substrate to room temperature, followed by a 90-minute primary incubation at 37°C, biotin-labeled detection antibody incubation for 60 minutes at 37°C, HRP-Streptavidin binding for 30 minutes at 37°C, TMB color development in the dark for 10-20 minutes, and measurement of absorbance at 450 nm after adding stop solution.
