Description

A colorimetric sandwich ELISA from Assay Genie designed for the quantitative measurement of IRE1 (Inositol-Requiring Enzyme 1, also known as ERN1) in rat-derived samples. The kit uses a double-antibody sandwich format with a sensitivity of 9.375 pg/mL and an assay range of 15.625–1000 pg/mL.

Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity for serum (n=5) is 87–96% at 1:2, 85–97% at 1:4, and 82–99% at 1:8; EDTA plasma (n=5) shows 85–100%, 84–96%, and 83–100%; and heparin plasma (n=5) shows 81–94%, 86–102%, and 92–99%. Spike-and-recovery averages are 97% for serum, 99% for EDTA plasma, and 94% for heparin plasma. Reagents are stored at 2–8°C for 12 months.

The assay workflow comprises reagent and TMB equilibration, a primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding for 60 minutes, HRP-Streptavidin incubation for 30 minutes, TMB color development for 10–20 minutes in the dark, and absorbance measurement at 450 nm after adding stop solution. IRE1 is a key enzyme in the unfolded protein response (UPR) and endoplasmic reticulum stress, with research relevance to diabetes, cancer, and neurodegenerative disorders.
