Rat Interferon-alpha Antibody/INF-alpha-Ab ELISA Kit

A sandwich ELISA, double-antigen format kit from Assay Genie for the quantitative detection of interferon-alpha antibody in rat samples. The assay is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples, and the manufacturer reports a sensitivity of 0.938 ng/mL with a working range of 1.563–100 ng/mL. Kits should be stored sealed at 2–8 °C and must not be cryopreserved.

Validation data supplied by the manufacturer show linearity across dilutions (1:2, 1:4 and 1:8; n = 5) for serum (86–96%, 86–96%, 86–104%), EDTA plasma (85–99%, 83–98%, 86–99%) and heparin plasma (88–96%, 81–97%, 82–94%). Spike-and-recovery performance is reported as 93–103% (average 97%) for serum, 92–105% (average 98%) for EDTA plasma and 87–100% (average 93%) for heparin plasma (n = 5 in each case).

The procedure uses a biotin-labelled antigen, an HRP–streptavidin conjugate and a TMB substrate, with absorbance read at 450 nm after a stop step. Recommended sample preparation includes clotting and centrifugation at 1000 × g for 20 minutes for serum, anticoagulant collection and centrifugation at 1000 × g for 15 minutes at 2–8 °C for plasma, centrifugation at 2500 rpm for 5 minutes for cell culture supernatant, and standard homogenisation or lysis for tissues and cells, with technical support available from the manufacturer for other sample types.
