The Assay Genie kit maps to the UniProt entry Q9WUK0 and detects INSL3 under several historical synonyms recorded for the same Leydig cell hormone, including RLNL, RLF, Relaxin-Like Factor and Leydig insulin-like peptide, which simplifies cross-referencing with earlier rodent reproductive literature that predates the unified INSL3 nomenclature.

Matrix validation reported by the manufacturer shows serum recovery of 80–102% (mean approximately 91%), EDTA plasma recovery of 81–100% (mean approximately 90%), and heparin plasma recovery of 80–89% (mean approximately 84%). Serial dilution linearity across 1:2 to 1:16 dilutions typically falls between 79% and 107%. Reproducibility is documented at intra-assay CV less than 10% and inter-assay CV less than 12%, and specificity testing showed no significant cross-reactivity with closely related relaxin-family analogues.

The assay workflow adds 100 µL of standard or sample per well for a 2 hour incubation at 37 °C, followed by 100 µL of prepared Detection Reagent A for 1 hour at 37 °C, three washes, 100 µL of Detection Reagent B for 1 hour at 37 °C, five washes, 90 µL of Substrate Solution for 15–25 minutes at 37 °C, and a final 50 µL of Stop Solution before immediate plate reading on a standard absorbance reader.

Unopened kits are validated for a 12 month shelf life from the date of manufacture, with measured activity loss below 5% across that window when stored and handled as directed. Common downstream applications include profiling circulating INSL3 in rodent models of cryptorchidism, gonadal dysgenesis, Leydig cell regeneration following chemical or surgical injury, ovarian theca cell function, and endocrine-disrupting chemical exposure, often alongside testosterone and luteinizing hormone readouts.
