The Assay Genie rat IGFBP3 ELISA kit uses a matched antibody pair format in which a pre-immobilized capture antibody binds IGFBP3 from the loaded sample, followed by sequential incubation with a biotinylated secondary antibody and streptavidin-HRP. Standards and unknown samples are added to antibody-coated microwells; after washing away unbound material, the detection antibody binds the captured analyte. HRP-TMB conversion produces a measurable chromogenic signal proportional to IGFBP3 concentration, and a standard curve generated from serial dilutions of the supplied reference standard is used to interpolate unknown values.

IGFBP3 is a 264-amino-acid glycoprotein that binds IGF-1 and IGF-2 with high affinity, modulating their interaction with cell-surface IGF receptors. Because circulating IGFBP3 concentrations change in response to GH status, fasting, and tumor-derived proteases such as PAPP-A, this kit is frequently used in studies of acromegaly, GH deficiency, type 2 diabetes, and cancer cell proliferation. Researchers working with rat models of metabolic syndrome, hepatic injury, or skeletal growth can analyze samples collected from tail-vein or trunk-blood draws. Tissue homogenates and cell lysates should be prepared in non-denaturing buffers with protease inhibitors to preserve the intact IGFBP3 complex.

Each kit provides reference standard, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer concentrate, sample diluent, TMB substrate, and stop solution. Allow all reagents to equilibrate to room temperature before use, and bring the TMB substrate to room temperature only immediately before addition to avoid background drift. Run samples in duplicate with appropriate dilution series when analyte concentrations are expected to fall near or above the upper limit of 10 ng/mL.
