Description

A colorimetric sandwich ELISA from Assay Genie is designed for the quantitative detection of insulin in rat samples. It uses a double-antibody sandwich format with a sensitivity of 46.875 pg/mL and a working range of 78.125-5000 pg/mL. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Insulin is a key hormone in regulating glucose metabolism, and changes in its levels can serve as a biomarker for conditions such as diabetes and obesity.

Validated linearity across 1:2, 1:4 and 1:8 dilutions falls within 87-105% for serum (n=5), 89-97% for EDTA plasma (n=5), and 91-104% for heparin plasma (n=5). Spike-and-recovery data show 85-103% (average 93%) for serum, 91-102% (average 97%) for EDTA plasma, and 85-103% (average 96%) for heparin plasma. The assay workflow includes reagent equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution. Reagents are stored at 2-8°C for 12 months.
