Description

Designed for the quantitative measurement of interleukin-7 in rat samples, this Sandwich ELISA Kit from Assay Genie uses a colorimetric, double-antibody detection method. The assay is reactive with rat samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types. IL-7 (Uniprot P56478), also referred to as Lymphopoietin-1 or PBGF, is a signalling molecule classified as a cytokine that plays a central role in T-cell development and immune system function.

Performance specifications include a sensitivity of 9.375 pg/mL and an assay range of 15.625–1000 pg/mL. Recovery values are reported for serum (range 87–100%, average 94%), EDTA plasma (range 83–100%, average 89%) and heparin plasma (range 86–94%, average 89%), with linearity values provided across 1:2, 1:4 and 1:8 dilutions for each matrix.

The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto pre-coated wells, and incubating at 37°C for 90 minutes to enable antigen binding. A biotin-labelled detection antibody is added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin for 30 minutes. TMB substrate is then added for 10–20 minutes in the dark, the reaction is stopped, and absorbance is measured at 450 nm. Sample-specific preparation guidance is supplied for serum, plasma, tissue homogenate, cell culture supernatant and cell lysates. The kit should be stored at 2–8°C for 12 months.
