The assay protocol follows six main steps: equilibrating reagents and TMB substrate to room temperature and setting up wells on the pre-coated plate; primary incubation of standards, samples and blanks at 37°C for 90 minutes; addition of biotin-labeled detection antibody with incubation at 37°C for 60 minutes; HRP-Streptavidin (SABC) binding at 37°C for 30 minutes; TMB color development in the dark for 10-20 minutes; and reading absorbance at 450 nm immediately after adding the stop solution.
