Rat IL-17 ELISA Kit

A colorimetric Sandwich ELISA, Double Antibody kit from Assay Genie for the quantitative measurement of rat IL-17 (Interleukin 17; synonyms include CTLA-8, IL17A, IL-17CTLA-8, IL17interleukin-17A, and interleukin 17A). The assay is reactive with rat samples.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 9.375 pg/mL and the standard curve range is 15.625–1000 pg/mL. Storage is 2–8°C for 12 months. The procedure involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and stop solution addition with absorbance measurement at 450 nm. Sample preparation guidance includes centrifuging clotted blood at 1000 × g for 20 minutes for serum, centrifuging anticoagulant-collected blood at 1000 × g for 15 minutes at 2–8°C for plasma, homogenizing tissue with PBS and protease inhibitors, centrifuging cell culture supernatant at 2500 rpm for 5 minutes, and lysing cells with appropriate buffer.

Reported linearity across dilutions 1:2, 1:4, and 1:8 is 85–102%, 84–99%, 81–93% for serum (n = 5), 85–101%, 86–99%, 81–95% for EDTA plasma (n = 5), and 86–105%, 84–98%, 84–99% for heparin plasma (n = 5). Spiked recovery is 93–104% (average 99%) for serum (n = 5), 85–105% (average 96%) for EDTA plasma (n = 5), and 87–105% (average 94%) for heparin plasma (n = 5).
