Description

A sandwich ELISA, double antibody kit from Assay Genie for the quantitative measurement of Heat Shock Factor 1 (HSF1) in rat samples. The assay accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and reports a sensitivity of 0.188 ng/mL with a standard range of 0.313-20 ng/mL. HSF1 is a key transcription factor in the cellular stress response, and its dysregulation has been associated with cancer, neurodegenerative disorders, and aging, so accurate quantification supports a range of research applications. Unopened kits are stored at 2-8°C for 12 months.

The procedure follows a standard sandwich workflow: equilibrate reagents and TMB substrate, load standards, samples and blanks onto the pre-coated plate, then incubate at 37°C for 90 minutes for antigen binding. After a wash, add biotin-labeled detection antibody and incubate at 37°C for 60 minutes, then add HRP-Streptavidin (SABC) for 30 minutes at 37°C. Develop colour with TMB substrate in the dark for 10–20 minutes, stop the reaction and read absorbance at 450 nm.

Reported spike-recovery averages from the manufacturer run between 93% and 95% across serum, EDTA plasma and heparin plasma, and dilution linearity (1:2 to 1:8) sits broadly in the 82–102% range across the same matrices.
