From a methodological standpoint, this kit uses a sandwich-style chemiluminescent immunoassay in which a monoclonal capture antibody pre-bound to the microplate wells binds rat HPA from loaded standards and samples, followed by incubation with a horseradish peroxidase-conjugated detection antibody and a luminol-based substrate. Signal generation is proportional to HPA concentration and is read on a luminometer or chemiluminescence-compatible plate reader. Typical run-time is approximately 3-4 hours at room temperature, with standards diluted in serial two-fold steps to construct an eight-point calibration curve spanning the working range, and at least two wells reserved for kit controls in each plate to monitor inter-assay consistency.

Sample handling guidance from Assay Genie recommends collecting serum or plasma with appropriate anticoagulants, clarifying by centrifugation at roughly 1000 x g for 10 minutes, and aliquoting into single-use vials before storage at -80 °C to limit freeze-thaw cycles that degrade HPA activity. Tissue homogenates should be prepared in cold PBS with protease inhibitors and clarified before loading. Investigations of glomerular basement membrane degradation in streptozotocin-induced diabetic rats, metastatic dissemination in syngeneic tumor models, and granulation tissue remodeling during dermal wound repair are all supported by this rat-specific CLIA when paired with parallel colorimetric ELISA for orthogonal validation.
