Description

Designed for rat research applications, this sandwich double-antibody ELISA from Assay Genie enables quantitative measurement of HMGB1 (High Mobility Group Box 1) protein in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit employs a colorimetric detection method with a sensitivity of 18.75 pg/mL and a measuring range of 31.25–2000 pg/mL, supporting accurate and precise detection of this key mediator of inflammation, cell differentiation, and immune response.

Elevated HMGB1 levels have been linked to diseases such as sepsis, arthritis, and cancer, making this kit relevant for studies into inflammatory and immune-related biological processes. The protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples, and controls onto the pre-coated microplate, incubating at 37°C for 90 minutes for antigen binding, then adding biotinylated detection antibody with a 60-minute incubation, HRP-Streptavidin (SABC) for 30 minutes, TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm immediately after stop solution. Reagents are stored at 2–8°C for up to 12 months.

Performance validation includes linearity data for serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with serum showing 87–103%, 85–97%, and 92–105%, EDTA plasma 85–101%, 85–98%, and 82–98%, and heparin plasma 80–96%, 80–85%, and 89–94%. Recovery ranges are reported as 87–104% (average 96%) for serum, 91–103% (average 99%) for EDTA plasma, and 89–101% (average 95%) for heparin plasma. The kit has been cited in published research, including a study on methotrexate-induced hepatotoxicity in rats.
