Description

From Assay Genie, this colorimetric Sandwich ELISA kit is intended for the quantitative detection of Hexosaminidase B (HEXB) in rat-derived specimens. Hexosaminidase B is an enzyme involved in lysosomal storage disorders that plays a key role in the degradation of glycosphingolipids, and its dysregulation has been implicated in conditions such as Tay-Sachs and Sandhoff disease, making the kit a useful tool for research into the pathogenesis of these disorders.

The double-antibody Sandwich ELISA accepts a wide range of sample types, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Manufacturer-reported performance characteristics include a sensitivity of 0.938 IU/mL and a measuring range of 1.563–100 IU/mL. Linearity has been demonstrated in serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, with reported ranges of 82–100%, 85–100% and 89–102% at the 1:2 dilution. Recovery ranges are 86–97% for serum, 85–102% for EDTA plasma and 86–103% for heparin plasma, with average recoveries of 98%, 96% and 93% respectively. The kit is intended for storage at 2–8°C for 12 months.

The assay procedure follows a stepwise incubation: equilibration of reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, colour development with TMB substrate for 10–20 minutes in the dark, and a stop reaction with absorbance measurement at 450 nm. Sample preparation guidance covers serum, plasma, tissue homogenate, cell culture supernatant and cell lysate.
