Description

A colorimetric Sandwich ELISA from Assay Genie designed for the quantitative measurement of hepcidin in rat samples. The kit uses a double-antibody sandwich format and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, making it broadly applicable to research in iron metabolism.

According to the manufacturer, hepcidin is a key regulator of iron homeostasis that controls the absorption and distribution of iron, with dysregulation linked to conditions such as anemia, hemochromatosis and inflammation. The assay reports a sensitivity of 9.375 pg/mL and a standard-curve range of 15.625-1000 pg/mL. Linearity for serum is 91-105% at 1:2, 85-103% at 1:4 and 86-101% at 1:8; EDTA plasma 82-99%, 91-98% and 84-100%; heparin plasma 83-99%, 83-98% and 81-97%. Recovery is reported as 85-102% (average 98%) for serum, 92-105% (average 98%) for EDTA plasma and 85-103% (average 94%) for heparin plasma. Reagents are stored at 2-8°C for 12 months.

The workflow involves equilibrating reagents and the TMB substrate, loading standards, samples and blanks onto the pre-coated plate, and incubating at 37°C for 90 minutes. A biotin-labelled detection antibody is then added for a 60-minute incubation, followed by HRP-Streptavidin for 30 minutes. TMB substrate is added and developed in the dark for 10-20 minutes, after which stop solution is introduced and absorbance is read at 450 nm.
