Description

This Sandwich ELISA from Assay Genie is designed for the quantitative measurement of Heparin Cofactor II (HC-II) in rat samples. Compatible specimen types include serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. The kit uses a double-antibody sandwich format with colorimetric readout and is positioned as a tool for research into blood coagulation regulation, with HC-II linked to thrombin inhibition and to conditions such as thrombosis and cardiovascular disease.

Reported performance characteristics include a sensitivity of 0.469 pmol/mL and an assay range of 0.781-50 pmol/mL. Linearity at 1:2, 1:4, and 1:8 dilutions showed ranges of 86-105%, 84-101%, and 81-100% for serum (n=5); 85-100%, 86-101%, and 93-100% for EDTA plasma (n=5); and 89-101%, 82-101%, and 82-99% for heparin plasma (n=5). Spike-and-recovery testing yielded ranges of 89-104% (average 94%) for serum, 85-102% (average 95%) for EDTA plasma, and 88-100% (average 94%) for heparin plasma. The kit is stored at 2-8°C for 12 months.

The protocol proceeds through reagent and plate equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB substrate development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution addition.
