Rat HDL/High-density lipoprotein ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of high-density lipoprotein (HDL) in rat samples, using a double-antibody sandwich format on a pre-coated microplate. Detection relies on a biotinylated antibody, an HRP-streptavidin conjugate, and a TMB substrate, with absorbance read at 450 nm following a stop reaction.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit reports a sensitivity of 1.875 ng/mL and a standard curve range of 3.125-200 ng/mL. Linearity in serum (n=5) was 87-104% at 1:2, 82-93% at 1:4, and 83-95% at 1:8; EDTA plasma (n=5) gave 86-99%, 83-101%, and 81-99% at the same dilutions, and heparin plasma (n=5) gave 88-105%, 87-96%, and 84-98%. Spike-and-recovery averaged 94% in serum (range 88-103%), 96% in EDTA plasma (range 87-105%), and 93% in heparin plasma (range 88-104%).

The protocol incubates the plate at 37°C — 90 minutes for antigen binding, 60 minutes with the biotinylated detection antibody, and 30 minutes with HRP-streptavidin — before TMB development in the dark for 10-20 minutes. Serum is clotted and centrifuged at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C. Unopened kit components are stored at 2-8°C and remain stable for 12 months under those conditions.
