Building on the workflow above, this 96-well microplate chemiluminescent immunoassay from Assay Genie uses a sandwich-format antibody pair directed against apolipoprotein A-I, the dominant protein component of the HDL particle, which ensures species-specific recognition in rat matrices without significant cross-reactivity to LDL or VLDL fractions.

Typical assay runtime is roughly 3.5 hours, beginning with 50 ul of pre-diluted serum or plasma added to each pre-coated well, followed by sequential incubations with the HRP-conjugated detection antibody and the luminol-based substrate solution. Signal is captured on a standard luminometer or glow chemiluminescence-capable microplate reader, and quantitation is performed against a seven-point calibrator curve calibrated to purified rat HDL.

The working dynamic range generally spans 0.5 to 100 ug/ml of HDL cholesterol equivalents, with intra-assay CV typically below 8% and inter-assay CV below 12%, and spike-and-recovery in rat serum matrices routinely falling between 85% and 115%. Samples reading above the highest standard should be diluted in the provided assay diluent and re-tested; plasma collected on EDTA or heparin is acceptable, while grossly hemolyzed or lipemic specimens may require additional clarification before testing.

Researchers commonly apply this kit in studies of high-fat diet-induced obesity, streptozotocin-treated diabetic rat models, statin and fibrate pharmacology, hepatic lipid export pathways, and atherosclerosis research using ApoE-knockout or Wistar rat backgrounds. For research use only, not for diagnostic procedures, and all components should remain refrigerated at 2 to 8 degrees C between uses, with the substrate reagent protected from light to preserve luminescent output across the full shelf life of the kit.
