Description

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of haptoglobin in rat samples using a double-antibody, colorimetric format. The kit is reactive with rat and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It targets haptoglobin (UniProt P06866), a protein associated with the immune response and inflammation, and which serves as a biomarker for conditions such as infection, inflammation and liver disease.

Reported performance characteristics include a sensitivity of 9.375 ng/mL and a standard curve range of 15.625–1000 ng/mL. Linearity (n = 5) for serum spans 85–105% at 1:2, 97–103% at 1:4 and 87–102% at 1:8; for EDTA plasma, 82–98% at 1:2, 85–98% at 1:4 and 83–99% at 1:8; and for heparin plasma, 86–96% at 1:2, 81–94% at 1:4 and 82–100% at 1:8. Spike-and-recovery (n = 5) gives mean values of 93% for serum, 99% for EDTA plasma and 95% for heparin plasma, with corresponding ranges of 85–100%, 86–105% and 90–101%. All components should be stored at 2–8°C and the kit is stable for 12 months under these conditions.

The workflow uses a biotin-labelled detection antibody with HRP–streptavidin (SABC) and TMB substrate. Standards, samples and blanks are incubated on the pre-coated plate at 37°C for 90 minutes, followed by 60 minutes with the detection antibody and 30 minutes with SABC. Colour is developed in the dark for 10–20 minutes, stopped, and absorbance is read at 450 nm. Sample preparation notes include allowing blood to clot and centrifuging serum at 1000 × g for 20 minutes, and collecting plasma at 1000 × g for 15 minutes at 2–8°C.
