Rat GUSB/beta glucuronidase ELISA Kit

A sandwich ELISA from Assay Genie designed for the accurate measurement of beta-glucuronidase levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Beta-glucuronidase is an enzyme that plays a crucial role in the metabolism of glucuronides, important in the detoxification and elimination of various substances from the body. Dysregulation of beta-glucuronidase levels has been linked to various diseases and conditions, making it a valuable biomarker for research applications. The kit is stored at 2-8°C for 12 months and uses a colorimetric detection method.

Performance characteristics include a sensitivity of 46.875 pg/mL and an assay range of 78.125-5000 pg/mL. Linearity at sample dilutions of 1:2, 1:4, and 1:8 was measured as follows: serum (n=5) at 87-98%, 91-99%, 86-105%; EDTA plasma (n=5) at 87-100%, 90-97%, 82-100%; and heparin plasma (n=5) at 80-90%, 81-98%, 83-100%. Recovery values average 99% (range 90-104%) for serum, 93% (range 89-100%) for EDTA plasma, and 96% (range 89-102%) for heparin plasma.

The protocol follows standard sandwich ELISA methodology: equilibrate reagents and TMB substrate to room temperature, then load standards, samples, and control wells on the pre-coated plate. Incubate at 37°C for 90 minutes for antigen binding. Add biotin-labeled detection antibody and incubate at 37°C for 60 minutes. Add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes. Develop with TMB substrate in the dark for 10-20 minutes. Stop the reaction and measure absorbance at 450 nm immediately.
