Description

Assay Genie supplies a sandwich ELISA, double-antibody kit for the quantitative measurement of Growth/differentiation factor 2 (GDF2) in rat-derived samples. The assay uses a colorimetric detection method and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Performance characteristics include a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Linearity testing at dilution ratios of 1:2, 1:4 and 1:8 (n=5) returned serum values of 89-104%, 85-98% and 82-94%; EDTA plasma values of 94-105%, 86-95% and 84-100%; and heparin plasma values of 87-105%, 87-98% and 81-94%. Spike-and-recovery testing (n=5) produced average recoveries of 95% for serum (range 90-99%), 96% for EDTA plasma (range 86-104%) and 98% for heparin plasma (range 91-104%).

The workflow has six steps: equilibrate reagents and TMB substrate to room temperature; incubate the pre-coated plate with standards and samples at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop colour with TMB substrate for 10–20 minutes in the dark; then add stop solution and read absorbance at 450 nm. Reagent components are stored at 2-8°C for up to 12 months. GDF2, also known as bone morphogenetic protein 9 (BMP-9), is implicated in bone formation and vascular development, and is relevant to research into skeletal disorders, cardiovascular disease and cancer.
