Rat Granzyme B ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of granzyme B in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody sandwich format with colorimetric detection, providing a sensitivity of 0.938 pg/mL across a working range of 1.563–100 pg/mL.

Granzyme B is a key enzyme associated with cytotoxic T cells and natural killer cells, where it contributes to target cell apoptosis and immune surveillance. Validated performance includes recovery of 86–104% in serum (average 97%), 95–104% in EDTA plasma (average 100%), and 95–103% in heparin plasma (average 100%), with linearity values ranging from 82–100% across the tested dilution factors (1:2, 1:4, 1:8) for serum, EDTA plasma, and heparin plasma samples. Components should be stored at 2–8°C and remain stable for 12 months under these conditions.

The standard protocol involves a 90-minute primary incubation at 37°C, followed by 60 minutes of biotin-labelled detection antibody binding, 30 minutes of HRP-streptavidin (SABC) binding, and TMB substrate development in the dark for 10–20 minutes, with absorbance read at 450 nm after addition of stop solution.
