Description

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of Glycogen Phosphorylase BB (GPBB/PYGB) in rat samples. The kit is intended for research applications in metabolism and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

The double-antibody sandwich format uses colorimetric detection via a biotin-labeled detection antibody and HRP-Streptavidin (SABC), with absorbance read at 450 nm. The sensitivity is 9.375 pg/mL and the assay range is 15.625-1000 pg/mL. Dilutional linearity at 1:2, 1:4 and 1:8 was reported for serum (85-101%, 84-100%, 85-96%), EDTA plasma (82-98%, 86-100%, 81-95%) and heparin plasma (85-104%, 85-105%, 86-102%). Spike-and-recovery averages were 92% for serum (range 86-101%), 98% for EDTA plasma (range 85-98%) and 97% for heparin plasma (range 87-105%).

The protocol involves reagent equilibration, a 37°C primary incubation for 90 minutes, detection antibody binding for 60 minutes at 37°C, HRP-Streptavidin binding for 30 minutes, TMB color development in the dark for 10-20 minutes, and stop solution addition before reading. Reagents are stored at 2-8°C for 12 months. GPBB is an enzyme involved in glycogen metabolism and the breakdown of glycogen into glucose, and abnormal levels have been linked to metabolic disorders, making it a useful biomarker in research.
