Description

A Sandwich ELISA from Assay Genie designed for the quantitative measurement of gp36/Podoplanin in rat samples. The kit uses a double-antibody, colorimetric detection method and is intended for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. According to the manufacturer, sensitivity is 9.375 pg/mL and the assay range is 15.625-1000 pg/mL. Reagents should be stored at 2-8°C for 12 months, and the kit is suited to cell biology research.

The procedure uses a biotin-labeled detection antibody and HRP-Streptavidin conjugate (SABC) with TMB substrate, and absorbance is read at 450 nm. Standard incubation steps include 90 minutes at 37°C for antigen binding, 60 minutes for detection antibody binding, and 30 minutes for SABC binding, followed by 10-20 minutes of color development. Reported linearity at 1:2, 1:4 and 1:8 dilutions ranges from 87-102%, 82-100% and 86-100% for serum (n=5), 90-102%, 83-98% and 81-88% for EDTA plasma (n=5), and 85-104%, 84-92% and 82-99% for heparin plasma (n=5). Recovery in serum averages 93% (range 88-97%), in EDTA plasma 95% (range 88-101%), and in heparin plasma 99% (range 89-105%).

GP36, also known as Podoplanin (UniProt Q64294), is involved in the regulation of cell migration, angiogenesis and metastasis, and is used as a biomarker in studies of cancer progression and lymphatic vessel formation.
