Rat Glycogen phosphorylase/PYGL ELISA Kit

This sandwich double-antibody ELISA from Assay Genie is designed for the quantitative measurement of Rat Glycogen phosphorylase/PYGL in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with kit storage at 2-8°C for 12 months. The microplate is pre-coated with a capture antibody specific to the target analyte, and signal is generated via a biotinylated detection antibody, HRP-conjugated Streptavidin, and TMB substrate, producing a colourimetric reaction read at 450 nm.

Linearity across 1:2, 1:4 and 1:8 dilutions was 86-104% / 82-99% / 80-89% for serum (n=5), 86-103% / 82-98% / 85-98% for EDTA plasma, and 88-100% / 87-99% / 81-100% for heparin plasma. Recovery ranges were 85-101% (serum, average 96%), 87-105% (EDTA plasma, average 94%) and 89-101% (heparin plasma, average 95%).

The standard protocol involves equilibrating reagents, loading standards and test samples onto the pre-coated plate and incubating at 37°C for 90 minutes, followed by incubation with biotin-labelled detection antibody at 37°C for 60 minutes, HRP-Streptavidin at 37°C for 30 minutes, TMB substrate development for 10-20 minutes in the dark, and stop solution prior to absorbance measurement at 450 nm.
