The Assay Genie Sandwich ELISA Kit enables quantitative measurement of Glutaminase (Gls), the kidney isoform of the mitochondrial enzyme that catalyzes the hydrolysis of glutamine into glutamate and ammonia. This colorimetric assay is formatted on a 96-well microplate and is validated for use with rat serum and plasma samples, making it well suited to research into renal ammoniagenesis, nitrogen metabolism, and glutamine-dependent bioenergetics in preclinical models.

The kit applies a sandwich immunoassay workflow with pre-coated wells, detection antibodies, and a chromogenic substrate that produces a measurable absorbance read at 450 nm on a standard microplate reader. Reconstituted reagents should be stored refrigerated at 2-8 degrees C, while unused strip wells should be kept sealed with desiccant to preserve immunoreactivity across the full plate. Researchers commonly pair this ELISA with western blot or PCR to cross-validate changes in Gls expression in studies of kidney physiology, metabolic acidosis, and tumor cell metabolism. For research use only.

Beyond renal physiology, mitochondrial glutaminase has emerged as a focal point in cancer metabolism research, where tumor cells frequently upregulate glutaminolysis to fuel proliferation. This rat-specific Assay Genie kit lets investigators quantify circulating and tissue-derived Gls levels with the throughput of a microwell plate format.

Each kit ships with a pre-coated 96-well plate, lyophilized recombinant standards, biotinylated detection antibody, streptavidin-HRP conjugate, TMB chromogenic substrate, stop solution, and a 20x concentrated wash buffer. The standard curve typically spans 0.31 to 20 ng/mL, with intra-assay and inter-assay CV values reported in the lot-specific certificate of analysis. Serum and plasma samples are commonly diluted 1:2 to 1:5 in the provided sample diluent before loading 100 µL per well in duplicate.

Bring all reagents to room temperature before use, and limit freeze-thaw cycles of the reconstituted standards to preserve antibody binding activity. Reconstituted components remain stable for up to 4 weeks when refrigerated at 2-8 degrees C; for longer studies, aliquot the detection antibody and freeze at -20 degrees C to extend shelf life up to 6 months. Investigators studying chronic kidney disease, hepatic encephalopathy, or the Warburg effect in rodent disease models often integrate this ELISA with western blot and quantitative PCR to triangulate transcriptional, translational, and systemic Gls changes. For research use only.
