Validation across matrices shows strong linearity: at 1:2, 1:4, and 1:8 dilutions, serum (n=5) spans 89-105%, 82-94%, and 84-95%; EDTA plasma (n=5) spans 93-101%, 83-98%, and 80-100%; heparin plasma (n=5) spans 90-101%, 82-101%, and 81-97%. Recovery averages are 94% for serum (range 85-105%), 93% for EDTA plasma (range 86-99%), and 91% for heparin plasma (range 85-101%).

Galectin-9 is implicated in immune regulation, inflammation, and cancer progression, and its expression has been linked to various diseases, supporting its use as a research biomarker. The workflow uses a biotin-labeled detection antibody with HRP-Streptavidin (SABC) signal amplification and TMB substrate, with absorbance read at 450 nm after a stop reaction.
