Description

Manufactured by Assay Genie, this sandwich double-antibody ELISA is designed for the quantitative measurement of Granulocyte Colony Stimulating Factor 3 (G-CSF) in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. G-CSF is a cytokine involved in the production and maturation of granulocytes, immune cells important for fighting infection and inflammation, and dysregulation of G-CSF has been implicated in cancer, inflammatory disorders, and autoimmune conditions.

The assay uses a colorimetric detection method with a sensitivity of 9.375 pg/mL and a measuring range of 15.625–1000 pg/mL. Reported linearity for serum (n=5) is 86–90% at 1:2, 83–99% at 1:4 and 83–101% at 1:8; for EDTA plasma (n=5) 85–99% at 1:2, 90–96% at 1:4 and 82–99% at 1:8; and for heparin plasma (n=5) 82–101% at 1:2, 87–99% at 1:4 and 86–99% at 1:8. Spike-and-recovery averages are reported as 97% for serum (range 88–105%), 96% for EDTA plasma (range 85–100%) and 95% for heparin plasma (range 92–103%). Reagents are stored at 2–8°C for 12 months.

The workflow involves equilibrating reagents to room temperature, loading standards and samples for a 90-minute primary incubation at 37°C, biotinylated detection antibody incubation for 60 minutes at 37°C, HRP-Streptavidin binding for 30 minutes at 37°C, TMB colour development for 10–20 minutes in the dark, and absorbance reading at 450 nm after stop solution addition. Serum is prepared by clotting and centrifugation at 1000 × g for 20 minutes; plasma by anticoagulant collection and centrifugation at 1000 × g for 15 minutes at 2–8°C.
