Description

A sandwich ELISA from Assay Genie designed for the quantification of G-6-Pase (Glucose-6-phosphatase), also referred to as G6PT and GSD, in rat samples using a double antibody format. G-6-Pase plays a central role in glucose metabolism by catalysing the final step in gluconeogenesis and glycogenolysis, and dysregulation of its activity has been linked to metabolic disorders such as diabetes and glycogen storage diseases, making it a valuable marker for research in metabolism. The kit is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples, and detection is colorimetric with absorbance read at 450 nm.

Reported performance characteristics include a sensitivity of 46.875 mIU/mL and a measuring range of 78.125–5000 mIU/mL, with kit stability at 2–8°C for 12 months. Linearity across 1:2, 1:4 and 1:8 dilutions is 85–102% / 84–98% / 81–94% for serum, 93–105% / 91–100% / 80–99% for EDTA plasma, and 85–103% / 82–98% / 80–100% for heparin plasma. Recovery is 86–101% (average 95%) for serum, 86–103% (average 92%) for EDTA plasma, and 86–104% (average 96%) for heparin plasma.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and a zero control onto the pre-coated plate, and incubating at 37°C for 90 minutes for antigen binding. A biotin-labelled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is added and incubated in the dark for 10–20 minutes, and absorbance is read at 450 nm immediately after the stop solution. Sample preparation guidance includes allowing blood to clot and centrifuging at 1000 × g for 20 minutes for serum, collecting plasma using anticoagulant tubes and centrifuging at 1000 × g for 15 minutes at 2–8°C, and homogenising tissue in PBS with protease inhibitors and collecting the supernatant after centrifugation for tissue homogenates.
