Spike-and-recovery in the actual matrix yielded 80-102% recovery in serum, 81-99% in EDTA plasma, and 80-89% in heparin plasma, with dilution linearity spanning 79% to 104% across 1:2 to 1:16 sample dilutions. The protocol layers cleanly onto a standard ELISA workflow: 50 µL of standard or sample plus 50 µL of Detection Reagent A incubate 1 hour at 37°C, followed by 100 µL of Detection Reagent B for another 1 hour at 37°C, then 90 µL of TMB Substrate Solution for 15-25 minutes at 37°C, terminated by 50 µL of Stop Solution. The complete microplate kit ships with a 12-month shelf life, and the supplied lyophilized standard as well as concentrated detection reagents tolerate multiple freeze-thaw cycles when aliquoted.
