Description

From Assay Genie, this Sandwich ELISA (Double Antibody) kit is intended for the quantitative measurement of fibromodulin (FMOD) in rat samples. The colourimetric assay has a stated sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL, and is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2–8°C for 12 months.

Fibromodulin is a protein involved in regulating collagen organisation and extracellular matrix structure, with roles in tissue development, wound healing and maintenance of tissue integrity. Dysregulation of fibromodulin has been linked to conditions such as arthritis, cardiovascular disease and fibrosis, making it a useful biomarker for related research. The assay uses a pre-coated dismountable microplate and follows a six-step protocol: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotin-labelled detection antibody binding at 37°C for 60 minutes; HRP-Streptavidin (SABC) binding at 37°C for 30 minutes; TMB colour development in the dark for 10–20 minutes; and a stop reaction with absorbance read at 450 nm.

Reported validation data for serum, EDTA plasma and heparin plasma (n = 5 per matrix) show linearity of 90–103%, 90–93% and 82–100% for serum; 89–99%, 86–96% and 80–91% for EDTA plasma; and 82–100%, 87–98% and 92–103% for heparin plasma at 1:2, 1:4 and 1:8 dilutions respectively. Recovery ranges were 89–102% (average 95%) for serum, 85–103% (average 98%) for EDTA plasma, and 88–100% (average 94%) for heparin plasma.
