Quantitative measurement of fibrinogen alpha chain (FGa) in rat plasma is supported by the Assay Genie ELISA Kit, a paired-antibody immunoassay formatted for 96 microplate determinations and read on a standard absorbance plate reader. Fibrinogen is a liver-synthesized soluble plasma glycoprotein, and circulating levels of its alpha subunit are commonly monitored in rat models of thrombosis, vascular injury, hepatic dysfunction, acute-phase inflammatory response, and metabolic disease.

The kit provides a complete reagent set: pre-coated microplate wells, matched antibody pair, calibrated protein standards spanning the working range of the assay, streptavidin-HRP detection reagents, TMB substrate, stop solution, and concentrated wash and dilution buffers. Each plate is sufficient for 96 individual wells run in duplicate against an eight-point standard curve, with capacity remaining for experimental samples and quality controls.

Citrate or EDTA-anticoagulated rat plasma is the validated sample matrix; plasma is separated by centrifugation, optionally aliquoted, and applied directly after dilution in the provided assay buffer. Workflow follows standard practice: standards, controls, and diluted plasma samples are added in duplicate, followed by biotinylated detection antibody and streptavidin-HRP conjugate. After incubation and washing, TMB substrate develops a colorimetric readout that is stopped with the included acid solution and read at the wavelength appropriate to the substrate. Sample FGa concentrations are then back-calculated against the standard curve to compare treatment and control cohorts in cardiovascular, pharmacologic, and toxicology studies conducted in rats.
