Rat FGF4 ELISA Kit

A high-quality Sandwich ELISA from Assay Genie designed for the quantitative measurement of fibroblast growth factor 4 (FGF4) in rat samples. The kit uses a double-antibody sandwich format and is validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Colorimetric readout is taken at 450 nm, with a reported sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL.

The protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks onto the pre-coated plate, incubating at 37°C for 90 minutes, adding a biotin-labelled detection antibody for 60 minutes, adding HRP-Streptavidin (SABC) for 30 minutes, developing with TMB substrate in the dark for 10–20 minutes, then adding stop solution and reading absorbance at 450 nm. Reported linearity for serum (n=5) is 88–99% at 1:2, 86–104% at 1:4 and 97–105% at 1:8; for EDTA plasma (n=5) 89–94%, 93–100% and 82–98%; and for heparin plasma (n=5) 80–93%, 83–100% and 84–99%. Recovery is 92–102% (average 99%) for serum, 86–101% (average 95%) for EDTA plasma and 87–101% (average 94%) for heparin plasma. Kit components are stored at 2–8°C for 12 months.

FGF4, also known as FGF-4, contributes to cell proliferation, differentiation and tissue development, and is involved in embryonic development, wound healing and tissue regeneration. Its dysregulation has been associated with cancer and developmental disorders, making it a useful biomarker for biomedical research (UniProt: F1LSQ8).
