Description

Manufactured by Assay Genie, this Sandwich ELISA Kit is designed for the precise quantitative measurement of ferritin heavy chain (Fth1) in rat-derived samples, supporting research into iron storage and homeostasis. It is for research use only and is reactive with rat samples, accepting serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay detects natural and recombinant rat ferritin heavy chain (UniProt P19132) and is categorised for the Neurosciences research area.

The assay delivers a detection range of 6.25-400 ng/mL with a sensitivity of 3.135 ng/mL. Reported precision is an intra-assay CV of 5.2% and an inter-assay CV of 9.7%. Linearity has been evaluated in serum (N=5) at dilutions 1:2, 1:4, 1:8 and 1:16, returning 109-119%, 107-116%, 103-112% and 98-107%; EDTA plasma (N=5) at the same dilutions gives 80-91%, 105-115%, 87-100% and 113-123%; and heparin plasma (N=5) gives 100-110%, 109-117%, 109-117% and 89-101%. Spike-and-recovery testing reports an average of 95% for serum (range 89-101%) and 97% for plasma (range 91-103%).

Ferritin heavy chain is a key subunit of the ferritin complex, which stores iron in a soluble, non-toxic and readily available form. It has ferroxidase activity, taking up ferrous iron and depositing it as ferric hydroxides, and contributes to iron delivery to cells, including uptake in capsule cells of the developing kidney. Dysregulation of ferritin levels has been implicated in iron overload disorders, inflammation and neurodegenerative conditions, providing a context in which this kit can be applied to study disease mechanisms and explore potential therapeutic strategies.
