Description

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of ferritin heavy chain in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody sandwich format with colorimetric detection and is positioned for cell biology research, including studies of iron storage and release, as well as conditions linked to dysregulated ferritin such as iron overload disorders, inflammation, and neurodegenerative disease.

The kit delivers a sensitivity of 3.75 ng/mL with a working range of 6.25-400 ng/mL. Reagents are stored at 2-8°C for 12 months, with lyophilised standard and microplate strips also suitable for -20°C storage up to 12 months. The supplied protocol covers reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB substrate development in the dark for 10-20 minutes, and absorbance reading at 450 nm after adding stop solution.

Linearity across 1:2, 1:4 and 1:8 dilutions (n=5) was 87-105% / 87-105% / 85-93% for serum, 88-100% / 87-101% / 82-96% for EDTA plasma, and 80-100% / 80-96% / 88-96% for heparin plasma. Spike-recovery ranges and averages were 85-99% (91%) for serum, 88-104% (93%) for EDTA plasma, and 96-105% (100%) for heparin plasma.
