Rat ERK2/MAPK1 ELISA Kit

Assay Genie offers a sandwich ELISA, double-antibody kit for the quantitative measurement of ERK2/MAPK1 in rat samples. Detection is colorimetric, with absorbance read at 450 nm after addition of TMB substrate and stop solution. The kit is intended for research into cell death, where ERK2/MAPK1 acts as a key signalling protein involved in cell growth, differentiation and survival, with dysregulation implicated in cancer, inflammatory disorders and neurological conditions.

The assay has a reported sensitivity of 0.469 ng/mL and a working range of 0.781-50 ng/mL. Dilutional linearity for serum (n=5) was 86-102% at 1:2, 92-101% at 1:4 and 89-103% at 1:8; EDTA plasma (n=5) gave 85-99%, 87-97% and 85-100% at the same dilutions; heparin plasma (n=5) gave 85-100%, 82-99% and 85-96%. Spike-and-recovery averaged 96% for serum (range 85-103%), 99% for EDTA plasma (range 85-104%) and 98% for heparin plasma (range 89-104%).

Accepted sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are stored at 2-8°C for 12 months, with several components protected from direct light. The procedure involves primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, and colour development in the dark for 10-20 minutes.
