From a research-use perspective, EDN1 in the rat is generated from preproendothelin-1 by endothelin-converting enzyme (ECE-1) and signals through two G-protein-coupled receptors, ETA and ETB, which mediate vasoconstriction in vascular smooth muscle and clearance in endothelial cells. Plasma EDN1 concentrations in rat models of salt-sensitive hypertension, deoxycorticosterone acetate (DOCA)-salt injury and streptozotocin-induced diabetes are often elevated two- to five-fold compared with normotensive controls, supporting its use as a biomarker of endothelial dysfunction in cardiovascular and renal research.

Sample handling matters: collect plasma into EDTA or aprotinin-containing tubes, centrifuge at approximately 1000 x g for 10 minutes within 30 minutes of draw, and store aliquots at -80 °C to preserve the peptide, which has a circulating half-life of only one to two minutes. Tissue homogenates from lung, kidney, heart and aorta should be lysed in the presence of protease inhibitors to prevent EDN1 degradation. The Assay Genie competitive format is well-matched to small-molecule peptide quantification where a sandwich approach is not feasible, and standard curves should be run in duplicate across the working range of 12.5 to 800 pg/mL with a sensitivity around 4.93 pg/mL.

Common applications include monitoring circulating EDN1 in pulmonary hypertension models, tracking renal cortical expression after ischemia-reperfusion injury, and quantifying peptide release from cultured rat endothelial cells, vascular smooth muscle cells or mesangial cells following stimulation with angiotensin II, thrombin or shear stress. Researchers should validate parallelism between their samples and the standard curve, since acidified plasma or urea-containing tissue extracts may require dilution into the assay buffer.
