Description

Manufactured by Assay Genie, this is a Sandwich ELISA, Double Antibody-based kit designed for the precise and reliable detection of dystrophin in rat samples. It is reactive with rat and accepts a broad range of liquid sample types, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The detection method is colorimetric, with absorbance read at 450 nm after a TMB substrate step. Dystrophin is a key protein involved in muscle function and structure, playing a crucial role in conditions such as muscular dystrophy, and the kit is positioned as a tool for researchers studying muscle health, disease progression, and therapeutic approaches in rat models.

Reported performance figures include a sensitivity of 0.469 ng/mL and a measuring range of 0.781-50 ng/mL. Linearity across dilutions (1:2, 1:4, 1:8; n = 5) is given as 85-104%, 83-94%, 85-98% for serum; 90-101%, 82-93%, 88-99% for EDTA plasma; and 89-100%, 84-93%, 81-97% for heparin plasma. Recovery (n = 5) is 90-97% (average 95%) for serum, 90-102% (average 99%) for EDTA plasma, and 88-99% (average 94%) for heparin plasma. The kit is intended to be stored at 2-8°C for 12 months. Components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody (100X), HRP-streptavidin conjugate SABC (100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer, and a technical manual.

The supplied assay procedure comprises six steps: reagent and plate preparation with standards, samples, and zero controls; primary incubation at 37°C for 90 minutes; biotin-labeled detection antibody incubation at 37°C for 60 minutes; HRP-streptavidin binding at 37°C for 30 minutes; TMB color development in the dark for 10-20 minutes; and a stop reaction with absorbance measurement at 450 nm. Sample preparation notes describe clotting/centrifugation for serum, anticoagulant collection and centrifugation for plasma, homogenization in PBS with protease inhibitors for tissue, clarification by centrifugation for cell culture supernatant, and lysis buffer treatment for cell lysates.
