Description
From Assay Genie, this Rat DPD/Deoxypyridinoline ELISA Kit is a competitive, antigen-coated enzyme-linked immunosorbent assay intended for research use. The microplate is pre-coated with the target analyte, and detection relies on a biotin-labelled antibody together with an HRP-streptavidin (SABC) signal system, with absorbance read at 450 nm.
Key specifications include a sensitivity of 1.875 ng/mL, a working range of 3.125–200 ng/mL, and reactivity with rat samples. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit should be stored sealed at 2–8°C and not cryopreserved. In validation testing, linearity across 1:2, 1:4 and 1:8 dilutions (n = 10) was 91–102% in serum, 87–105% in EDTA plasma and 87–105% in heparin plasma; spike-and-recovery averaged 94% for serum, 93% for EDTA plasma and 92% for heparin plasma.
The assay workflow involves adding 50 µL of standard or sample together with 50 µL of biotin-labelled antibody per well and incubating for 45 minutes at 37°C, adding SABC working solution for 30 minutes at 37°C, developing with TMB substrate for 10–20 minutes in the dark, and stopping the reaction before reading at 450 nm. Deoxypyridinoline is described by the manufacturer as a biomarker of bone turnover and collagen breakdown, supporting research into osteoporosis, arthritis and metabolic bone disease in preclinical rat models.
