Description

Manufactured by Assay Genie, this Rat Disabled homolog 2 (Dab2) ELISA Kit is a sandwich, double-antibody enzyme-linked immunosorbent assay for the quantitative measurement of rat Disabled homolog 2 in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. Detection is colorimetric, using a biotin-conjugated antibody followed by avidin conjugated to horseradish peroxidase (HRP) and the substrate TMB, with absorbance read at 450 nm ± 2 nm after addition of stop solution.

The kit is specific for natural and recombinant rat Disabled homolog 2, with a stated detection range of 0.156–10 ng/mL and a sensitivity of 0.083 ng/mL. Disabled homolog 2 (Dab2), also referred to as Adaptor molecule disabled-2, DOC-2, Doc2, C9, Mitogen-responsive phosphoprotein and Differentially expressed in ovarian carcinoma 2 (UniProt O88797), is described as a clathrin-associated sorting adaptor required for clathrin-mediated endocytosis of cargoes containing non-phosphorylated NPXY internalisation motifs such as the LDL receptor. The protein is also reported to participate in integrin and TGF-beta receptor signalling, in modulation of Wnt/beta-catenin and androgen receptor signalling, and has been described as a candidate tumour suppressor.

Disabled homolog 2 localises to the cytoplasm, clathrin-coated pits, clathrin-coated vesicle membrane and the plasma membrane. The kit is shipped on ice packs, is intended for research use only, and includes validated Bradford reagent for total protein quantification and sample normalisation. Storage requirements and the expiry date are indicated on the kit box label.
