Rat DIO1/Deiodinase, Iodothyronine, Type I ELISA Kit

This colorimetric Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of rat Type I iodothyronine deiodinase (DIO1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Using a double antibody detection format, the assay provides a sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL, supporting reproducible results for research in metabolism, thyroid hormone biology, and metabolic disorders.

Validated linearity data demonstrate consistent performance across dilutions. In serum (n=5), recoveries ranged from 85–100% at 1:2, 84–101% at 1:4, and 81–98% at 1:8. EDTA plasma (n=5) showed 90–105%, 82–96%, and 81–97% at 1:2, 1:4, and 1:8 respectively, while heparin plasma (n=5) gave 87–102%, 82–98%, and 80–100% at the same dilutions. Spike-and-recovery testing returned 96–101% (average 98%) for serum, 85–101% (average 91%) for EDTA plasma, and 85–103% (average 92%) for heparin plasma.

The protocol follows a standard sandwich workflow: equilibrate reagents and TMB substrate to room temperature, load standards, samples and controls onto the pre-coated plate and incubate at 37°C for 90 minutes, add the biotin-labelled detection antibody and incubate at 37°C for 60 minutes, add HRP-Streptavidin and incubate at 37°C for 30 minutes, develop with TMB substrate in the dark for 10–20 minutes, then add stop solution and read absorbance at 450 nm. All components are stored at 2–8°C for 12 months. DIO1 is a key enzyme responsible for the conversion of thyroid hormones, playing a vital role in metabolic regulation and thyroid hormone homeostasis, with dysregulation implicated in obesity, diabetes, and thyroid dysfunction.
