Rat DCN (Decorin) ELISA Kit

Assay Genie's decorin (DCN) ELISA kit is a sandwich-format, double-antibody immunoassay developed for the quantitative measurement of decorin in rat-derived samples. Decorin is a small leucine-rich proteoglycan of the extracellular matrix that contributes to collagen fibrillogenesis, cell adhesion, and growth factor binding, and its dysregulation has been associated with fibrosis, cancer, and inflammatory disease, making it a relevant target for biomarker and pathogenesis studies.

The assay uses a colorimetric detection method. Manufacturer-reported performance includes a sensitivity of 4.688 pg/mL and a working range of 7.813-500 pg/mL. The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with recommended storage at 2-8°C for 12 months. The protocol involves equilibrating reagents to room temperature, loading standards, samples and a blank on the pre-coated plate and incubating at 37°C for 90 minutes, adding biotin-labelled detection antibody and incubating at 37°C for 60 minutes, adding HRP-Streptavidin (SABC) and incubating at 37°C for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution.

Reported linearity at 1:2, 1:4 and 1:8 dilutions spans 85-99% to 86-103% for serum, 83-95% to 84-101% for EDTA plasma, and 82-99% to 80-91% for heparin plasma. Recovery ranges are 91-94% (average 93%) for serum, 90-100% (average 94%) for EDTA plasma, and 89-105% (average 96%) for heparin plasma. The kit is supplied with a dismountable ELISA microplate, lyophilised standard, biotin-labelled detection antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, and a plate sealer.
