Citrate synthetase is an enzyme that plays a critical role in the citric acid cycle, a key metabolic pathway in cellular energy production, and dysregulation of its activity has been implicated in various metabolic disorders and diseases, making it a valuable biomarker for metabolism-related research.

The kit demonstrates a sensitivity of 1.875 ng/mL with an assay range of 3.125-200 ng/mL. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and components should be stored at 2-8°C for 12 months. Reported linearity spans 86-105% for serum, 84-100% for EDTA plasma, and 81-100% for heparin plasma across 1:2, 1:4, and 1:8 dilutions (n=5). Recovery ranges are 88-98% for serum (average 93%), 90-101% for EDTA plasma (average 96%), and 90-103% for heparin plasma (average 96%).

The detection method is colorimetric, with absorbance read at 450 nm following TMB substrate development. The assay procedure involves reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes, HRP-Streptavidin (SABC) binding for 30 minutes, and color development in the dark for 10-20 minutes before stop solution addition.
