Description

This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of Cross Linked C-telopeptide of Type II Collagen (CTX-II) in rat samples. CTX-II is a key biomarker associated with cartilage degradation and is widely used to study the progression of joint diseases such as osteoarthritis, as well as to evaluate potential therapeutic interventions in preclinical models. The assay uses a double-antibody sandwich format with colorimetric detection and is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Linearity has been validated in serum (85–103% at 1:2, 91–104% at 1:4, 86–99% at 1:8; n=5), EDTA plasma (82–101% / 83–97% / 82–101%; n=5), and heparin plasma (87–99% / 84–91% / 83–100%; n=5). Recovery ranges from 94–105% in serum (n=5, average 99%), 86–97% in EDTA plasma (n=5, average 92%), and 90–101% in heparin plasma (n=5, average 96%). Reagents should be stored at 2–8°C for 12 months.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature and setting standard, test sample and control (zero) wells on the pre-coated plate; a 90-minute primary incubation at 37°C; a 60-minute incubation with biotin-labelled detection antibody; a 30-minute HRP-Streptavidin binding step; TMB substrate development for 10–20 minutes in the dark; and absorbance measurement at 450 nm immediately after addition of stop solution.
