Description
From Assay Genie, this sandwich, double-antibody colorimetric ELISA is designed for the quantitative measurement of Cartilage Oligomeric Matrix Protein (COMP) in rat samples. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.
Reported performance characteristics include a sensitivity of 0.938 ng/mL and an assay range of 1.563–100 ng/mL, with reagents stored at 2–8°C for up to 12 months. Dilution linearity in serum, EDTA plasma and heparin plasma samples tested at 1:2, 1:4 and 1:8 (n = 5) ranged from 80–105%. Spike-and-recovery across the same matrices fell within 85–105%, with average recoveries between 93% and 98%.
The protocol comprises a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-Streptavidin (SABC) binding step, TMB colour development in the dark for 10–20 minutes, and an absorbance read at 450 nm after the addition of stop solution. According to Assay Genie, COMP is a key protein in cartilage and other connective tissues with a role in joint health, and altered COMP levels have been linked to osteoarthritis and rheumatoid arthritis, supporting its use as a biomarker in musculoskeletal disease research.
