Rat COL4 (Collagen Type 4) CLIA Kit (RTES00139)

A chemiluminescence immunoassay from Assay Genie is designed for the quantitative detection of collagen type 4 (COL4) in rat samples. The sandwich-format kit is suitable for use with serum, plasma and other biological fluids, using a sample volume of 100 μL, and the protocol takes approximately 4.5 hours to complete.

The assay has a sensitivity of 56.25 pg/mL and a detection range of 93.75–6000 pg/mL. No significant cross-reactivity or interference between Rat COL4 and analogues was observed, and both intra-assay and inter-assay CV values are reported as < 15%. Recovery has been validated across serum (range 95-108%, average 101%), EDTA plasma (range 93-110%, average 101%) and cell culture media (range 88-98%, average 93%).

The micro CLIA plate is pre-coated with antibody specific to Rat COL4. Standards or samples are added to the wells and combined with the specific antibody. A biotinylated detection antibody specific for Rat COL4 and an Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each well and incubated. Free components are washed away and the substrate solution is added. The relative light unit (RLU) value is measured by a chemiluminescence immunoassay analyzer; RLU is positively associated with the concentration of Rat COL4, which is calculated by comparing sample RLU values to the standard curve.
