Rat COL2/CollagenII ELISA Kit

Assay Genie offers a sandwich ELISA, double antibody assay intended for the quantitative measurement of collagen type II (COL2) in rat-derived samples. The kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and components are stored at 2-8°C for 12 months.

Manufacturer-specified performance characteristics include a sensitivity of 37.5 pg/mL and an assay range of 62.5-4000 pg/mL, with absorbance read at 450 nm after TMB colour development. Reported linearity for 1:2, 1:4 and 1:8 dilutions is 85-105% for serum, 87-100% for EDTA plasma and 90-105% for heparin plasma (n=5 for each). Reported recovery averages 91% for serum, 93% for EDTA plasma and 98% for heparin plasma, with corresponding recovery ranges of 88-97%, 85-104% and 95-103%. Detection is colorimetric, using a biotin-labelled detection antibody with HRP-streptavidin (SABC) and TMB substrate.

According to Assay Genie, collagen type II is a key structural protein of cartilage, and changes in its levels have been linked to joint disorders, making the kit applicable to research in cartilage biology and joint disease. The protocol described involves reagent equilibration, sample addition with incubation at 37°C for 90 minutes, biotinylated detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin incubation at 37°C for 30 minutes, and TMB incubation in the dark for 10-20 minutes before stopping the reaction.
